Primary "target-positive" hybridoma clones are archived and the secreted antibodies are analyzed for their performance in specific applications. Once the leads are identified, we carry the primary clones through a subcloning and validation process. These final clones are archived as eight validated subclones from the parental line.
We generate antibody from hybridomas, transiently and stably transfected cells, in serum-free media. Our typical threshold for antibody production is 1000mg of research-grade and preclinical material depending on the method and capacity of the cells to generate antibody. We typically purify antibody using protein A and protein G columns on our AKTA pure HPLC.
As well as purifying monoclonal antibodies, we perform analysis on samples through size exclusion chromatography typically using a Superdex 200 Increase 10/300 GL. Size exclusion chromatography will provide information about a sample’s purity as well as information about militarization. This can be used in conjunction with our protein production services or independently.
